The tourists elected to field first at Cape Town's Newlands - the famous Test match venue sitting at the foot of Table Mountain. South Africa made 259 for eight from their 50 overs, with Norfolk star Mickleburgh claiming two for 33 from his six overs.
Batting at number five, Mickleburgh entered the fray with England on 53 in the 18th over, joining Derbyshire's Daniel Redfern at the crease.
The pair added 32 in the next eight overs and Mickleburgh looked to be finding his feet when he struck his first boundary in the 26th over. But three balls later he was caught and bowled by Stephan Smith, having made 17 from 28 balls.
Mickleburgh's Essex team-mate Adam Wheater briefly threatened a late revival as he made a brisk 30, but England were all out for 209 in the 45th over.
The second one-day international of the five-match series takes place today at Boland Bank Park, Paarl.
